Im Jahr 2414 wird bei einer Bohrung nach Ressourcen auf dem Planeten Aegis VII ein "Marker" gefunden. Dieser wird von der Sekte der Unitologen, die sich nach dem Fund des ersten Markers vor 200 Jahren gründeten, als heilig angesehen. Das Bergbauschiff Ishimura nimmt den Marker an Bord. Daraufhin mehren sich Gewalt und psychische Störungen und der Marker enthüllt sein entsetzliches Geheimnis. --- Der Film ist Teil einer Serie, die durch Comics, Videospiele und Filme erzählt wird. Mit Comics werden die vorgelagerten Ereignisse auf Aegis VII erzählt, wo Kolonisten durch den Marker infiziert werden. Zeitgleich zum Film zeigt das Spiel DS Extraction das Schicksal von 4 Personen auf der Ishimura. Das Spiel Dead Space schließt direkt an diesen Film an, wo man mit einer Rettungsmission auf die Ishimura kommt. Man entdeckt, dass der Marker vor langer Zeit von den Unitologen gebaut wurde und auf die Erde gebracht werden sollte, bringt ihn zurück auf die Oberfläche und flieht mit einem Shuttle.
